What is FOXO4-DRI

FOXO4-DRI is a synthetic D-retro-inverso peptide designed to disrupt the interaction between the FOXO4 protein and p53 in senescent cells. By displacing p53 from the nucleus, it induces selective apoptosis in these 'zombie' cells, which accumulate with age and secrete inflammatory factors (SASP). The seminal work of Baar et al. (Cell, 2017) demonstrated restoration of fur density, renal function, and physical fitness in mice, establishing it as a prototype senolytic.
