What is MK-677 (Ibutamoren)

MK-677 (Ibutamoren) is an oral, non-peptide GH secretagogue that acts as a ghrelin receptor (GHSR) agonist, increasing pulsatile release of growth hormone and IGF-1 without suppressing endogenous production. Being orally active and having a long half-life, it allows once-daily dosing. Clinical trials (Nass et al., Ann Intern Med 2008) showed sustained increases in GH/IGF-1 and lean mass gain in the elderly.

Side effects

•Increased appetite, fluid retention, edema, paresthesias, lethargy, and elevated blood glucose/insulin resistance. May raise cortisol and prolactin in some cases.

Contraindications

•Diabetes or significant insulin resistance, heart failure, active cancer, pregnancy, and lactation. Caution with fluid retention and carpal tunnel syndrome.
