What is Retatrutide
Retatrutide is a unique triple agonist that simultaneously activates GIP, GLP-1, and glucagon receptors, combining appetite suppression with increased energy expenditure via the glucagon component. In the phase II study published in the New England Journal of Medicine (2023), participants achieved a mean weight loss of approximately 24% at the 12 mg dose over 48 weeks. It is in development by Eli Lilly in the phase III TRIUMPH program.

- •Dose-dependent gastrointestinal effects are the most common: nausea, diarrhea, vomiting, and constipation, generally mild to moderate and transient. An increase in heart rate related to the glucagon component may occur.
Contraindications
- •Contraindicated in people with a personal or family history of medullary thyroid carcinoma or multiple endocrine neoplasia type 2 (MEN2). Not recommended during pregnancy, lactation, or in active pancreatitis.
