What is Tesamorelin

Tesamorelin is a stabilized analog of human GHRH (1-44) that stimulates the pituitary gland to release endogenous GH in a pulsatile manner. It was approved by the FDA in 2010 (Egrifta) for the reduction of abdominal lipodystrophy in patients with HIV, based on phase III trials that demonstrated a significant reduction in visceral adipose tissue. By preserving the physiology of the GH-IGF-1 axis, it avoids the supraphysiological spikes of exogenous GH.

Side effects

  • •Injection-site reactions (erythema, pruritus), arthralgia, myalgia, peripheral edema, paresthesias, and elevated IGF-1. It may increase blood glucose and reduce insulin sensitivity.

Contraindications

  • •Pregnancy, active malignancy, hypothalamic-pituitary axis dysfunction, hypersensitivity to mannitol. Use caution in diabetics and in retinopathy.
